Functional Analysis of Anti-yeast Substance Secreted from Rahnella aquatilis AY2000 Strains for Antioxidant and Antihistamine Activity

Abstract
Reactive oxygen species (ROS) possess highly reactive response to macromolecules such as carbohydrate, nucleic acid, protein and amino acid in organism. Thus, various materials have been developed by researchers to remove ROS. So far, anti-oxidant material was purified from lots of plants and was applied for the production of health care products or skin care goods. In present research, an anti-yeast substance (AYS) purified from Rahnella aquatilis strain anti-yeast (AY) 2000 was verified for the ability of anti-oxidant and anti-histamine. H2O2 was scavenged by 21.2% and 20.3% with 0.02 and 0.04% AYS concentration, respectively. Especially, H2O2 was removed by 55.7% with 0.08% AYS concentration. In addition, AYS showed the effect of cell protection on HaCaT keratinocyte exposed to 0.25 mM H2O2. Furthermore, 0.005¡­0.08% AYS had an excellent scavenging effect against OH radical in dose-dependent pattern. Moreover, we examined the effect of AYS on the compound 48/80-induced histamine release in human mast cell (HMC)-1. The culture supernatants were harvested, and under 1¡­100 ¥ìg/ml concentrations, AYS inhibited histamine release in HMC-1 stimulated with compound 48/80 (300 ¥ìg/ml). These results indicated that AYS might be an useful skin care agent for anti-fungus as well as anti-oxidant and anti-histamine.
